摘要
目的研究代谢型谷氨酸受体、谷氨酸转运体在糖尿病脑损伤时的基因表达情况及作用机制。方法 Wis-tar大鼠28只,随机分成A(常规饲料喂养组,n=14)、B(高糖高脂饲料喂养+STZ注射诱导胰岛素抵抗,即模型组,n=14)二组,自由进食水,12h光照周期。饲养一个月后,测定血糖。6个月后以RT-PCR检测糖尿病鼠脑皮质EAAT-1、mGluR-4、mGluR-6的mRNA表达情况。结果 B组mGluR-6 mRNA表达与A组相比明显增强(P<0.05),而EAAT-1、mGluR-4 mRNA表达与A组相比略有增强。结论 mGluR-6增高可能参与糖尿病脑组织细胞损伤。
Objective the purposes of this test is to study gene expression and mechanism concerning receptors of metabolic pattern(transporter of aminoglutaminic acid) in brain injured with diabetes.Methods There are twenty-eight Wistar rats to be randomly distributed class of A(this is control group breeding with routine animal feeds,the numbers are fourteen.) and B(this is test group breeding with high glucose and high fat animal feeds,in addition to inducing insulin resistance through injectioned STZ,the numbers are fourteen.).Drink and food of all rats is random,and receiving12 hours illumination cycle.A month later,all rats are be detected blood glucose.Six months later,the mRNA expression of EAAT-1、mGluR-4、mGluR-6 is be monitored with RT-PCR in pallium of diabetes' rats.Results the mRNA expression of mGluR-6 in class of B is more obviously increase than A(P0.05),however,the mRNA expressions of EAAT-1 and mGluR-4 is poor compared with class of A.Conclusion the increase of mGluR-6 may play a part in brain injured with diabetes.
