摘要
目的将流式细胞术测定酪氨酸磷酸化STAT3的方法用于外周血白细胞内酪氨酸磷酸化STAT3的检测,为JAK—STAT途径以及其他细胞内信号转导分子的研究和临床检测提供新的实验手段。方法建立酪氨酸磷酸化STAT3的流式细胞检潮方法,收集正常人及白血病患者的外周血标本,经磷酸化STAT3FITC标记后,用流式细胞术进行检测。结果在11例白血病患者的外周血标本中,检测出2倒标本有STAT3酪氨酸(Y705)磷酸化活化。结论部分白血病患者外周血白细胞中有酪氨酸(Y705)磷酸化STAT3表达。FCM检测方法可用于外周血标本STAT3的检测。
Objective To certify the abnormal express and activate of STAT3 in human leukemia cells,detected the tyrosine phosphorylated of STAT3 in white blood cells(WBC) of human peripheral blood by flow cytometry. Methods Detected tyrosine phosphorylated STAT3 in WBC of peripheral blood in normal group and patients with leukemia,using flow cytometry. Results Detected tyrosine (Y705) phosphorylated STAT3 in 2 peripheral blood samples among samples from 11 patients with leukemia. Conclusion There were tyrosine(Y705) phosphorylated STAT3 expression WBC of peripheral blood of partial patients with leukemia. Application of flow cytometry should be valuable in screening for signaling defection in selected patients with leukemia,cancers and so on. In addition,this technique should permit dissection of a full range of cellular signaling pathways at the protein level.
