摘要
目的:建立高效液相色谱法(HPLC法)测定昆明山海棠搽剂体外透皮吸收方法并研究制剂中月桂氮花卓酮的最佳用量。方法:以HPLC法测定雷公藤内酯醇(以下简称甲素)含量。结果:回归方程为Y=0.3265X+0.0266(r=0.9986,n=5)。日内RSD为0.5%(n=5);日间RSD为1.2%(n=5);方法的平均回收率为(92～98)%,RSD≤1.4%(n=5)。月桂氮花卓酮的最佳用量为2.0%,稳态透皮速率J=0.46μg·cm-1·h-1。结论:HPLC法简单易行,可用于甲素含量测定和透皮吸收研究。
OBJECTIVE:To establish HPLC method to assay for permeability in vitro through excised rat skin of triptolide in the Liniment of T.hypoglacum (Lev1.)Hutch ,and to decide an amount of azone.METHODS:The concentration of triptolide was determined by HPLC method. RESULTS:The results showed that the linear relationship was obtained between the concentration of triptolide and the peak area ration in the range of ( 10.0 ～ 100.0 ) μg·ml -1 ( r= 0.998 6 ,n=5 ).The RSD of within day were 0.5% ( n =5),and between day 1.2% ( n =5).The mean recovery were (92～98)%, RSD≤ 1.4% (n=5) .The experiment showed that the quantity of azone were 2.0% in the best, J =0.46 μg·cm -2 ·ml -1 .CONCLUSION:The HPLC method is simple,it was suitable to detect the concentration of triptolide and to study the permeability of the Liniment in vitro.
