摘要
目的:检测nm 23 H1 与m uc1 基因在乳腺癌中的表达水平,探讨nm 23 H1 与m uc1 在乳腺癌原发灶与转移灶中表达水平改变的临床意义. 方法:采用定量 R T P C R 与图像扫描定量分析技术检测22 例家族性乳腺癌及60 例散发性乳腺癌原发灶与转移灶中nm 23 H1 与 m uc1 基因的表达水平.结果:①在22 例有家族史的原发灶中, nm 23 H1 基因在10 例无转移的标本中检出率为8/10,12 例转移淋巴结中检出率4/12,癌旁组织中检出率为14/22;60 例散发性乳腺癌中,nm 23 H1 基因在40 例无转移的原发灶中检出率为 34/40,20 例转移淋巴结中检出率为7/20,癌旁组织中检出率为39/60;无转移的原发灶与转移灶中的检出率存在显著性差异( P< 001);②nm 23 H1 基因在乳腺癌原发灶呈高表达,在转移灶中呈低表达,表达量在两者之间存在显著性差别( P< 0.001);nm 23 H1 基因表达水平的高低与乳癌的分级、病理类型、家族史无关;③ m uc1 在22 例有家族史的无转移原发灶中检出率为2/10,转移淋巴结中检出率为10/12,癌旁组织中检出率为 15/22;60
AIM: To investigate the alteration of expression levels of nm23 H1 and muc1 in primary breast cancer and metastatic cancer, and to explore further the clinical significance of alteration of expression levels of nm23 H1 and muc1 . METHODS: By using quantitative RT PCR and Imagine Scanning Quantification Analysis System to examine the expression levels of nm23 H1 and muc1 in 22 samples of breast carcinoma with family history and 60 samples of dispersed breast carcinomas. RESULTS:①In 22 specimens of primary breast cancers with heredity family history, positive rates of nm23 H1 were respectively 8 /10 in primary breast cancers without metastasis, 4/12 in metastasis nodes, 14/22 in paracancer tissues ; in 60 specimens of dispersed breast cancers, positive rates of nm23 H1 were respectively 34/40 in primary breast cancers without metastasis,7/20 in metastasis nodes, 39/60 in paracancer tissues;there were significantly differences between positive rates of nm23 H1 in primary tumors and ones in metastasis tumors ( P< 0.01); ②Expressional level of nm23 H1 was higher in primary breast cancers than in metastatic tissues, there was a significant difference between expressional levels in primary breast cancer and ones in metastatic tumors ( P< 0.01); expressional levels of nm23 H1 had no correlated with degrees of cancers and pathological types and heredity family history; ③In 22 specimens of primary breast cancers with heredity family history, rates of muc1 were respectively 2 /10 in primary breast cancers without metastasis, 10/12 in metastatic nodes, 15/22 in paracancer tissues ; in 60 specimens of dispersed breast cancers, rates of muc1 were respectively 10/40 in primary breast cancers without metastasis, 16/20 in metastatic nodes, 45/60 in paracancer tissues;there were significant differences between rates of muc1 in primary tumors and ones in metastatic tumor
