兔VX2瘤脊柱转移动物模型的实验研究 被引量:4

Metastatic tumor of VX2 carcinoma in rabbit spine: model establishment and experimental study

摘要 目的通过模拟脊柱转移性肿瘤的动脉血行转移机制建立兔VX2瘤脊柱转移动物模型,研究其生物学特性。方法新西兰大白兔共20只,随机平均分为A组和B组。均采用VX2肿瘤细胞悬液经腹主动脉短时阻断注射法建立椎体高选择性脊柱转移肿瘤模型。接种肿瘤后每天观察兔下肢神经功能变化,接种术后第2周起每周行CT影像学检查肿瘤生长情况。动物出现下肢瘫痪、死亡或接种后6周为观察终点,取出病椎并作组织学检查。结果 A组成瘤率70%;B组成瘤率80%。观察期内A组中2只出现完全瘫痪,3只不全瘫,5只未出现神经功能障碍;B组中3只出现完全瘫痪,3只不全瘫,4只未出现神经功能障碍。结论经腹主动脉短时阻断注射法建立兔VX2瘤脊柱转移动物模型具有高选择性,使肿瘤细胞富集于目标椎体,能较好地模拟肿瘤细胞经动脉系统移行至脊柱并形成转移灶的病理过程,对于探索脊柱转移肿瘤的发病机制具有重要意义。 Objective To establish the spinal tumor model of VX2 carcinoma in rabbits by simulating the mechanism of metastatic tumor through arterial system and observe its biological feature.Methods Twenty New Zealand white rabbits were randomly divided into 2 equal groups(Group A and B).The 2 experimental groups were injected respectively with different dosages of VX2 carcinoma cell suspensions into abdominal aorta,the low level of which was shortly occluded at the same time.The motor function of lower limbs was assessed daily after injection and CT scan was taken weekly after the 2nd week of post-injection.Observation was kept on 6 weeks unless the onset of paralysis or death occurred,and the pathological exam was performed when the observation ended.Results The tumor formation rate of Group A was 70%,and 80% of Group B,In Group A,complete and incomplete paralysis of the lower limbs occurred in 2 and 3 rabbits respectively,and 5 rabbits remained neurologically normal;in Group B,complete and incomplete paralysis of the lower limbs occurred in 3 and 3 rabbits respectively,and 4 rabbits remained neurologically normal.Conclusion Open trans-abdominal aorta injection of tumor cell is a feasible method to establish spinal metastatic tumor model of VX2 carcinoma in rabbits.This model has the similar metastatic mechanism of the trans-arterial system and progression with the natural process that tumor lesions develop to compress spinal cord and lead to paralysis in clinical cases of spinal metastasis,which will be helpful for the study on various treatment protocols.
