羌活醇提取物对肝缺血再灌注损伤大鼠肝组织抗氧化酶的影响 被引量:10

Effect of Alcohol Extract of Qianghuo(Notopterygium Root or Rhizome) on the Antioxidase of Liver Tissues of Rats with Hepatic Ischemia Reperfusion Injury

摘要 目的探讨羌活醇提取物对肝缺血再灌注损伤(HIRI)大鼠模型抗氧化作用的影响及机制。方法将100只大鼠随机分为空白组、模型组、丹参预处理组(DS组)和羌活预处理组(QH组)4组,后3组组内再分为缺血45min(I45)和缺血45min再灌注30min(I45R30)、缺血45min再灌注60min(I45R60)3个时间点,DS组、QH组分别给予丹参醇提取物、羌活醇提取物灌胃,均1ml/d,连续7d。行HIRI造模后分别测定各组大鼠各时点即刻血清丙氨酸氨基转移酶(ALT)水平和肝组织丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-Px)活力水平。结果模型组、QH组和DS组内大鼠模型I45R30、I45R60时点的血清ALT水平、MDA含量、GSH-Px活力均较I45时点明显升高(P<0.01);DS组I45R60时点大鼠肝组织中MDA含量较QH组明显降低(P<0.05);DS组I45R60时点大鼠肝组织GSH-Px活力均较模型组和QH组低(P<0.05)。结论羌活醇提取物可能是通过提高HIRI模型大鼠肝组织GSH-Px活力,增强对氧自由基的清除能力,减轻脂质过氧化损伤,从而起到保护肝脏的作用。 Objective To study the effect of alcohol extract of Qianghuo(Notopterygium root or rhizome)on antioxidase of liver tissues of rats with hepatic ischemia reperfusion injury(HIRI)and its mechanism.Methods Totally 100 rats were randomized into four groups,i.e.blank group,model group,Danshen(Salvia miltiorrhiza)pretreatment group(DS group),and Qianghuo(Notopterygium root or rhizome)pretreatment group(QH group).The latter three groups were subdivided into three groups respectively,namely,ischemia 45 min subgroup(I45),ischemia 45 min reperfusion 30 min group(I45R30)and 60 min subgroups(I45R60).The alcohol extract of Danshen or Qianghuo was given to DS group or QH group respectively by gastric perfusion 1 ml/d for 7 days.The level of alanine transaminase(ALT)in serum,content of malondialdehyde(MDA),and activity of glutathione peroxidase(GSH-Px)of liver tissues of all rats in each time point were determined respectively and compared.Results In the I45R30 and I45R60 subgroups of model group,QH group,and DS group,the level of ALT in serum,the content of MDA,and the activity of GSH-Px were significantly higher than those of I45 subgroup(P0.01).In the I45R60 subgroup of DS group,the content of MDA of liver tissues was significantly lower than that QH group(P0.05).In the I45R60 subgroup of DS group,the activity of GSH-Px of liver tissues was lower than that of the model group and QH group(P0.05).Conclusion Qianghuo(Notopterygium root or rhizome)probably protects the liver through enhancing the GSH-Px activity,strengthening the ability of eliminating radical,and relieving the lipid peroxidation injury of liver tissue of HIRI model rats.
