摘要
目的:研究褐藻多糖硫酸酯(FPS)抗肾间质纤维化的作用机制。方法:采用不同浓度的孵育体外培养的人肾间质成纤维细胞(HRIF),观察24h、48h时HRIF增殖与凋亡的情况,检测纤维连接蛋白(FN)、层黏连蛋白(LN)、转化生长因子-β1(TGF-β1)的分泌水平。结果:FPS孵育HRIF24h、48h均可显著抑制HRIF的增殖(P<0.01);FPS可显著减少HRIF对FN、LN的分泌(P<0.01),而对TGF-β1的分泌差异无统计学意义(P>0.05);FPS对体外培养的HRIF细胞无诱导凋亡作用。结论:FPS可明显抑制体外培养的HRIF增殖,而且可以减少FN、LN两种细胞外基质的分泌,这可能是FPS抗肾间质纤维化、防治慢性肾衰竭的作用机制之一。
Objective:To reveal the mechanism of action that fucoidan counteracts renal interstitium fibrosis.Methods:FPS in different concentration was used to foster the cultured human renal interstitial fibroblasts(HRIF)in vitro to observe its proliferation and apoptosis and to detect the secretion of fibronectin(FN),laminin(LN),and transforming growth factor β1(TGF-β1)at the time points of 24 h and 48 h.Results:FPS inhibited proliferation of HRIF significantly(P0.01).FPS inhibited the expression of FN and LN(P0.01),without the effect on expression of TGF-β1(P0.05).Detected by flow cytometry,apoptotic peak of HRIF which were incubated with FPS for 24 hours was not find.Conclusion:It was shown that fucoidan significantly inhibited proliferation of human renal interstitial fibroblast,reduced the secretion of FN,LN.Fucoidan may delay the development of renal interstitium fibrosis,this may be the mechanism of action that FPS counteracts renal interstitium fibrosis.
