摘要
参照杨连甲的方法从小牛骨基质中提取牛骨形成蛋白(bBMP),经SDS-PAGE电泳分析,bBMP含67、43、30、22、18和14kD六种蛋白组分,保留BMP活性蛋白成份,植入小鼠股部肌肉后的X线及组织学观察发现,所提取的bBMP具有较高的骨诱导活性,bBMP植入后14天即可见有新骨形成,21天时有含骨髓的板层状骨形成。采用bBMP单克隆抗体的免疫组织化学染色发现,BMP存在于未分化间充质细胞胞浆和骨形成过程中的成骨细胞内。
Bovine bone morphogenetic protein(bBMP) was extracted from cortical bone matrix of one_year_old steers according to the procedure reported by Yang. The bBMP showed the same electrophoretic pattern as that reported in SDS polyacrylamide gel electrophoresis. The molecular weight of bBMP was determined to be 67on of bBMP, and lamellated new bones with marrow cavities were found on the 21th day of implantation. BMP/McAb was used for demonstration of BMP in BMP_bone inducing process by the immunohistochemical method. BMP was found in the cellular plasma of the un_differentiated mesenchmal cells and in osteoblasts during the stage of bone formation.
