摘要
目的研究并建立逍遥散(柴胡、白芍、当归、白术、茯苓、甘草、薄荷和炮姜)的高效液相色谱指纹图谱,为其质量控制提供可靠方法。方法色谱柱Venusil XBP C18(250 mm×4.6 mm,5μm);柱温35℃;体积流量1.0 mL/min;检测波长280 nm;进样量10μL;乙腈-0.2%磷酸水溶液梯度洗脱。结果逍遥散中多数峰可以达到基线分离。用中药色谱指纹图谱相似度评价系统对10批样品的指纹图谱进行峰匹配,确定22个共有峰,10批逍遥散中共有峰的相对保留时间RSD小于1%,指纹图谱的相似度均在0.99以上,选取共有峰在单味药材中均找到归属,柴胡、茯苓、白术中没有共有峰。结论该法建立的指纹图谱灵敏度高,稳定性好,可以做为逍遥散的质量控制手段并为其有效成分的研究提供依据。
AIM To provide a sensitive method for evaluation of Xiaoyao Powder(Bupleuri Radix,Paeoniae Radix alba,Angelicae sinesis Radix,Atractylodis macrocephalae Rhizoma,Poria,Glycyrrhizae Radix et Rhizoma,Menthae haplocalycis Herba and Zingiberis Rhizoma praeparatum) by HPLC fingerprint.METHODS Venusil XBP C18 column(250 mm×4.6 mm,5 μm) was used with the column temperature at 35 ℃;the detection wavelength was set at 280 nm,and the injection volume was 5 μL at 1.0 mL/min flow rate and a gradient solution of acetonitrile and H2O(acidified to 0.2% with phosphoric acid).RESULTS Most peaks could be separated preferably,and the RSD of the relative relation time of ten batches of Xiaoyao Powder was lower than 1%,twenty-two common peaks were obtained in the fingerprint of ten batches of Xiaoyao Powder with the help of peaks match with Similarity Evaluation System for Chromatographic Fingerprint of TCM.The similarity among fingerprints was calculated more than 0.99.The common peaks selected belonged to all the ingredients that could be used to classify the species,but Bupleuri Radix,Poria and Atractylodis macrocephalae Rhizoma did not share common peaks.CONCLUSION The fingerprinting could be used as a method of quality control and provides evidence for the research of effective components of Xiaoyao Powder.
