摘要
以枯草芽孢杆菌为试验菌株,采用液态培养基发酵,探讨枯草芽孢杆菌产α-淀粉酶的最佳发酵条件,分别对碳源、氮源、发酵时间、接种量、培养基初始pH进行单因素试验,在此基础上对碳源浓度、氮源浓度、接种量、培养基初始pH这4个因素进行了L9(34)正交优化试验。结果表明,葡萄糖优于其他碳源,尿素优于其他氮源,最佳培养时间为30 h;最佳发酵参数组合为pH 5.0,葡萄糖质量浓度0.2%,尿素质量浓度1.5%,接种量2.5%。
Singal factor and orthogonal analysis were applied to obtain the optimal conditions for fermenting α-amylase by Bacillus subtilis in liquid culture medium.Five factors,carbon source,nitrogen source,original pH,incubation time and inoculum size were tested respectively;and based on these single factor tests,the carbon concentration,nitrogen concentration,original pH value and inoculum size were analyzed in L9(34) orthogonal analysis.The result showed that the optimal carbon source and nitrogen source were glucose and carbamide,respectively.The optimal incubation time was 30 h.The optimal parameter was original pH value,5.0;carbon concentration,0.2%;nitrogen concentration,1.5%;and inoculum size,2.5%.
