摘要
目的通过动物实验,研究应用外源性的rhBMP-2后牵引成骨区生长因子TGF-β1的表达。方法在32只成年大耳白兔的一侧下颌骨前部行骨切开术,分别将空白胶原、rhBMP-2胶原复合物植入下颌骨切开处,用牵引器延长一侧下颌骨4 mm,分别于牵张完成后的第1、3、7、14 d处死一组动物,取牵引区新生骨痂行组织学及TGF-β1免疫组化染色,通过细胞图像分析仪测量牵引区新骨生成的平均灰度值,利用SPSS软件采用方差分析方法进行统计分析。结果牵引区周围组织均愈合良好,牵引间隙新骨逐渐形成,应用rhBMP-2组中的新骨组织周围TGF-β1的表达比空白胶原组中的多。结论动物实验表明,应用外源性的rhBMP-2后牵引成骨区生长因子TGF-β1的表达明显增强。
Objective To study the expression and function of TGF -β1 in the rabbit's mandibular distraction osteogenesis process. Methods Unilateral mandibular osteotomies were performed in 32 mature rabbits, rhBMP - 2 with the collagen carrier were implanted in the osteotomy side of mandibles. Only collagen sponge was implanted in control group. The mandibles of 48 rabbits were lengthened by 4mm with a distractor. At the 1, 3, 7, 14 days after the end of distrac- tion all animals were killed and the distracted calluses were harvested and processed for histological and immunohistochemistry study of TGF - β1. The average gray values of distracted calluses were analyzed by Cell digital Imaging Software. Finally the changes were analyzed by SPSS with the method of ANOVA. Results During the distraction period, there was no problem with the animals and the distractors. All the distractors and the around tissues were obtained good healing. The regenerated bone was found in the distraction gap after madibular lengthening by radiography and histology. The mandibular side treated with rhBMP - 2 had greater expressions of TGF - β1than it was treated with collagen sponge. Conclusion rhBMP - 2 could accelerate bone formation of mandibular distraction osteogenesis in rabbit, and could increase the expression of TGF - β1
