摘要
目的:研究端粒酶抑制剂3′-叠氮3′-脱氧胸腺核苷(AZT)对SGC 7901胃癌细胞凋亡及Bax蛋白和增殖细胞核抗原(PCNA)表达的影响,探讨其诱导肿瘤细胞凋亡的可能机制。方法:将处于对数生长期的SGC 7901胃癌细胞分为对照组及1.0 mmol.L-1AZT处理组,应用AZT 6 d后,采用MG-P-MY组合染色法检测SGC 7901胃癌细胞凋亡率,采用免疫组织化学EnVision法检测SGC 7901胃癌细胞Bax蛋白及PCNA的表达。结果:AZT组及对照组SGC 7901胃癌细胞凋亡率分别为(16.421±0.713)%和(3.366±0.271)%,AZT处理组SGC 7901胃癌细胞凋亡率明显高于对照组(P<0.05);对照组Bax蛋白及PCNA阳性细胞表达率分别为(16.851±10.064)%和(63.332±22.767)%,AZT处理组Bax蛋白及PCNA阳性细胞表达率为(47.389±18.042)%和(23.446±13.055)%,与对照组比较,AZT处理组Bax蛋白表达率明显增加(P<0.05),PCNA表达率明显降低(P<0.05)。结论:AZT能促进SGC 7901胃癌细胞Bax蛋白的表达,抑制PCNA表达,增加SGC 7901胃癌细胞凋亡。
Objective To study the influence of 3′-azido-3′-deoxythymidine(AZT) on the apoptosis of gastric cancer cell line SCG 7901 and the expressions of BAX protein and proliferation cell nuclear antigen(PCNA) and explore the mechanisms.Methods The gastric cancer cell line SCG 7901 treated with 1.0 mmol·L-1 AZT for 6 d were used as AZT group,at the same time,control group was set up.The apoptotic rates of SGC 7901 in two groups were detected with MG-P-MY staining.The expressions of Bax protein and PCNA of gastric cancer cell line SCG 7901 6 d after drug administration were determined by immunohistochemical technique(EnVision).Results The apoptotic rates in AZT proup and control group were(16.421±0.713)% and(3.366±0.271)% respectively,the apoptotic rate in AZT group was significantly higher than that in control group(P0.05).Compared with control group,the expression of Bax protein was significantly increased(16.851%±10.064% vs 47.389%±18.042%,P0.05) and the expression of PCNA was significantly decreased(23.446%±13.055% vs 62.332±22.767%,P0.05) in AZT group.Conclusion AZT can induce the apoptosis of gastric cancer cell line SCG 7901,promote the expression of Bax protein and inhibit the expression of PCNA.
