兔激素性股骨头缺血性坏死髋关节的骨质差异

摘要 目的建立兔激素性股骨头缺血性坏死模型,探讨兔激素性股骨头缺血性坏死髋关节周围骨质的差异。方法选取14只新西兰大白兔,雌雄各半。使用马血清加醋酸泼尼松龙制备兔激素性股骨头缺血性坏死模型,于激素注射完毕6周后,检测兔股骨头、小转子、髋臼的空骨陷窝率、骨髓的脂肪细胞直径与面积,用免疫组化法和W estern-b lot检测股骨头、小转子、髋臼骨质中VEGF的表达。结果实验动物的股骨头、小转子、髋臼的空骨陷窝率、骨髓脂肪细胞面积与直径均高于正常对照组;实验组中股骨头组、小转子组、髋臼组三组间在空骨陷窝率(F=131.125,P<0.05)、骨髓脂肪细胞面积百分比(F=41.114,P<0.05)、骨髓脂肪细胞直径(F=34.232,P<0.05)、骨组织中VEGF的表达均有统计学意义;股骨头组与小转子组在骨髓脂肪细胞面积百分比间的差异无统计学意义(P=0.131,P>0.05)。结论激素对兔髋关节周围骨质具有一定的影响,并且对股骨头、小转子、髋臼骨质的影响具有差异性。 Objective To establish the steroid-induced avascular necrosis of the femoral head in rabbits and to investigate the difference of bone around the hip-joint.Methods Fourteen adult rabbits(male,n= 7;female,n= 7).The horse serum combining prednisolone acetate was used to establish the animal model of steroid-induced avascular necrosis of femoral head.Six weeks after the injection,the histopathology was used to examine the hip-joint,and the bone marrow fat cells were examined morphologically.The immunohistochemistry and Western-Blot was used to detect VEGF in the hip-joint.Results In the femoral head group,lesser trochanter group and acetabula group was higher than that in normal group with the empty bone lacuna rate,bone marrow fat cells area and diameter.In the experiment group,there was difference between the empty bone lacuna rate,bone marrow fat cells area and diameter and relative concentration of VEGF.Conclusion The study shows the corticosteroid hormone has influence in hip-joint,and this effect is different in three groups.
