赤霉素对球等鞭金藻3011生物量、细胞形态与不同生长时期群体细胞活性的影响 被引量:3

摘要 目的观察赤霉素(gibberellins,GA)的添加量对球等鞭金藻3011(Isochrysis galbana 3011)生物量、细胞形态与不同生长时期群体细胞活性的影响。方法取处于指数生长期的球等鞭金藻3011藻液,分别添加终浓度为0(溶剂对照)、0.001、0.003、0.005、0.007、0.01、0.03、0.05、0.07、0.1 mg/L的赤霉素溶液,测定球等鞭金藻3011的密度和细胞形态;采用流式细胞技术对指数生长期(第8天)、相对生长下降期(第13天)和静止期(第18天)的球等鞭金藻3011群体细胞活性进行测定。结果添加0.005 mg/L赤霉素可使球等鞭金藻3011的生物量达到最高(1.76×108/ml);且与溶剂对照组比较,球等鞭金藻3011细胞面积增大,亮度提高(P<0.05或P<0.01)。0.07 mg/L赤霉素可使相对生长下降期群体细胞的活性最高(99.73%),引起指数生长期延长。结论 0.005 mg/L赤霉素可促进球等鞭金藻3011生物量和细胞形态指数的增加;0.07 mg/L赤霉素可提高群体细胞活性,延长了指数生长期。 Objective To observe the effect gibberellins (GA) content on the biomass,cell morphology and population activity with different growth stages of Isochrysis galbana 3011. Methods Isochrysis galbana 3011 was divided into control group and nine experimental groups. The density and cell morphology were determined by micro imaging morphology in exponential growth phase; the population activity was contrasted in exponential growth,relative growth reduction and stationary phase at the 8th, 13th and 18th day respectively in the same quantity with the method of flow cytometry affected by GA at the exposure dose of 0,0.001,0.003,0.005,0.007,0.01,0.03,0.05,0.07 and 0.1 mg/L. Results Biomass extreme was 1.76×10^8 cells/ml when GA was 0.005 mg/L, area and average brightness were significantly higher compared with the other groups (P〈0.05 or P〈0.01). When GA was 0.07 mg/L,population activity was reached to 99.73% as GA was 0.07 mg/L in relative growth reduction phase, exponential growth phase was extended. Conclusion At the GA exposure dose of 0.005 mg/L,cell morphological index of lsochrysis galbana 3011 will increase, and as GA is at 0.07 mg/L,the population activity will be enhanced, the exponential growth phase be prolonged.
