摘要
为研究补阳还五汤抗脑缺血的有效成分部位及其配伍关系,从补阳还五汤中提取了7 类有效成分部位,采用小鼠双侧颈总动脉和迷走神经结扎法造成脑缺血模型,研究各有效成分部位的作用,以正交实验法分析各有效成分部位的交互作用及有效剂量组合。结果:总生物碱、总多糖、总甙、总甙元和挥发油单独应用均可使小鼠耐缺血时间明显延长,而蛋白质和氨基酸单独应用无明显作用;正交实验表明,蛋白质、氨基酸与其它有效部位之间以及两者之间均无交互作用;三水平正交实验表明,在配伍上,各有效成分部位应取的有效剂量组合为:生物碱为生药0.58 g/kg 体重,多糖为生药11.44 g/kg 体重,甙元为生药8.74 g/kg 体重,总甙为生药14.4 g/kg 体重,挥发油为生药2.5 g/kg 体重。结论:在补阳还五汤中,具有抗脑缺血作用的有效成分部位主要为生物碱、多糖、总甙、甙元和挥发油,它们按一定的比例配伍可发挥较好的作用。
Object:Available compositions abstracted from Buyanghuanwu decoction and their compatibilities on antagonisming experimental cerebral ischemia.Method:Seven available compositions were abstracted from Buyanghuanwu decoction. Effects of each available composition on cerebral ischemia in mice induced by ligating bilateral common carotid artery and Vagus were studied.Interaction and the combination of effective dose of each composition were studied with orthogonal test.Result:1)Total alkaloids,Polysaccharides,saponins,aglycones and volatiles applied alone could prolong obviously the time of ischemia tolerance,but proteins and amino acids applied alone had no effect on the time of ischemia tolerance.2)Orthogonal test showed that there was no interaction between proteins and amino acids or other compositions.3)Tree levelsorthogognal test showed that the effective doses of each available composition were as follows:Alkaloids,0.58 g crude drug/kg;Polysaccharides,11.44 g crude drug/kg;Aglycones,8.74 g crude drug/kg;Total saponins,14.4 g crude drug/kg;Volatiles,2.5 g crude drug/kg.Conclutions:In Buyanghuanwu decoction,Alkaloids,polysaccharinges,total saponins,aglycones and volatiles are main available compositions in antogonisming cerebral ischemia.Compatibility of the available compositions according to definite proportion may play the most role in antagonisming cerebral ischemia in mice.
