摘要
目的初步筛选髓母细胞瘤中差异表达显著的微RNA(miRNA)并探求其生物学功能。方法应用基因芯片获得差异表达的miRNAs,对其中差异显著的目的miRNA行实时定量PCR验证,再利用生物信息学方法预测其潜在靶基因以及靶基因所富集的生物学通路。结果 miR-130a在髓母细胞瘤中显著下调,生物信息学分析表明,血小板衍生的生长因子受体α(PDGFRA)为其靶基因,且该基因与Ras/MAPK通路有关。结论 Ras/MAPK通路在髓母细胞瘤中上调,可能是miR-130a下调引起PDGFRA上调所致。
Objective To find the significant differentially expressed microRNAs in medulloblastoma and investigate their biological function.Methods Differentially expressed microRNAs were found by gene chips,and miR-130a was selected and verified using real-time quantitative PCR,then its potential target genes and their enriched biological pathways were predicted by bioinformatics softwares.Results miR-130a was down-regulated obviously in medulloblastoma.Bioinformatics analysis indicated that its target gene PDGFRA was involved in Ras/MAPK signaling pathway.Conclusion Down-regulation of miR-130a induced the up-regulation of PDGFRA,which might be the reason for up-regulation of Ras/MAPK signaling pathway in medulloblastoma.
