摘要
目的:提取小鼠GV(germinal vesicle,GV)期卵母细胞并分析其核质比。方法:用挤压法从卵巢中分离获得GV期卵母细胞,放入DEPC-NS液体快速清除颗粒细胞,获得GV期卵母细胞裸卵,并计算其核质比。结果:用挤压法分离获得的GV期卵母细胞有一完整球形核,核膜明显,含有单个折光的核仁,胞浆中心稍变黑并呈颗粒状,共51枚,测得其卵径为(40.940±3.341)μm,核径为(15.020±2.236)μm,其核质比为0.054±0.019。结论:用挤压法加DEPC-NS液体可快速破坏颗粒细胞之间的连接,便于迅速获得GV期卵母细胞裸卵;小鼠GV期卵母细胞的核质比为0.054±0.019,与其它物种同时相核质比在相同范围内,提示核质比是卵母细胞发育成熟的重要指标。
Objective:To extract the GV-stage oocytes of mouse and analyzed its the nucleus-plasma ratio.Methods: GV-stage oocytes were extracted and isolated by extrusion.Granulosa cells were quickly removed in DEPC-NS liquid and denuded oocytes were obtained.The nucleus-plasma ratio of the denuded oocytes was caculated.Results: The GV-stage oocytes had complete spherical nucleus with obvious nuclear membrane,and a single refractive nucleolus.Its cytoplasm center was a little black and in granular.A total of 51 GV-stage eggs were obtained,which were(40.940±3.341) μm in diameter and(15.020±2.236) μm in core diameter,and the nucleus-plasma ratio was 0.054 ± 0.019.Conclusions: The extrusion method and DEPC-NS liquid can rapidly destruct the connection between the granulosa cells,in which the denuded GV-stage oocytes are easy to obtain.The nucleus-plasma ratio in the mice GV-stage oocytes is 0.054 ± 0.019,while is similar to other reports.It suggests that the nucleus-plasma ratio is an important indicator of oocyte maturation.
