摘要
目的研究同型半胱氨酸(Hcy)对脑微血管内皮细胞(BMEC)损伤后NF-κB及其下游调控因子细胞间黏附分子1(ICAM 1)的影响。方法通过培养大鼠BMEC,用1 mmol/L浓度的Hcy及CuSO_4μmol/L共同干预培养的BMEC,分别作用0、6、12和18h,观察不同时间点细胞NF-κB p65及ICAM-1的表达。结果与0h比较,用Hcy干预6 h后,细胞开始出现ICAM-1蛋白表达,但差异无统计学意义(P>0.05),至1 2 h ICAM 1蛋白表达明显升高(P<0.05),18 h升高更明显(P<0.01);与0h比较,Hcy干预6h后,细胞NF-κB p65表达明显增加(P<0.05),至12h NF-κB p65核转位更明显(P<0.01);而18h NF-κB核转位的现象开始减弱,差异无统计学意义(P>0.05)。结论 Hcy可以促进NF-κB的核转位,诱导ICAM-1的表达。
Objective To study the influence of homocysteine(Hcy) on NF-κB p65 and ICAM-1 in cultured brain microvascular endothelial cells(BMEC). Methods Cultured BMEC were exposed to 1 mmol/L Hcy and CuSO4 (4μmol/L)for 0 h, 6 h, 12 h and 18 h. NF-κB p65 and ICAM-1 levels were evaluated by means of immunohistochemistry,FCM and Western blot. Results After BMEC were exposed to 1 mmol/L Hcy plus 4μmol/L CuSO4 for 12 h,the expression of ICAM-1 was significantly increased (P 〈0. 05) compared with 0 h exposure, especially after exposure for 18 hours (P 〈0.01). At the same time,the nuclear transfer of NF-κB p65 was peaked at 12 h (P 0. 01) and then decreased gradually(at 18 h, P〉 0.05). Conclusion Hcy could lead to the nuclear transfer of NF-κB p65 and the expression of ICAM-1 in a time-dependent manner.
