端粒酶逆转录酶高表达新生鼠真皮细胞株的构建及其生物学功能的初步研究 被引量:1

摘要 目的:以逆转录病毒载体pLEGFP-N1-TERT为介导构建TERT高表达新生鼠真皮细胞株,并观察其相关生物学功能变化情况。方法:将pLEGFP-N1-TERT感染C57新生鼠真皮细胞,并用G418筛选法进行阳性克隆的筛选并培养观察,同时将空载对照。两组细胞均经MTT法检测生长情况,流式检测表面标记物CD44、CD133的表达情况,Westernblot法检测TERT蛋白的表达,免疫组化检测Versican表达。结果:荧光显微镜下观察TERT高表达细胞发出绿色荧光,其生长速度较对照趋缓,其CD44、CD133、TERT蛋白、Versican表达均升高。结论:本实验获得TERT高表达新生鼠真皮细胞株,其毛发诱导能力较高,可作为毛囊细胞移植种子细胞来源之一。
