摘要
目的:研究慢性胰腺炎(CP)大鼠胃肠运动功能的变化及马来酸曲美布汀对CP的治疗效果。方法:健康Wistar大鼠30只,随机分为对照组、慢性胰腺炎组(模型组)和马来酸曲美布汀组(治疗组),每组10只。治疗组给予马来酸曲美布汀溶液灌胃,剂量为0.2mL/kg,1次/d;模型组及对照组灌注等量的生理盐水,3组均灌注2周。采用酶联免疫吸附(ELISA)法测定血清胆囊收缩素(CCK)水平,采用硝酸还原酶法测定血清NO水平;检测各组小肠运动系数、胃窦及小肠环行平滑肌肌条收缩能力、环行平滑肌细胞收缩反应;测定胃肠肌层诱导型一氧化氮合酶(iNOS)mRNA的表达量。结果:模型组小肠运动系数、胃窦和小肠环行平滑肌肌条的收缩力、平滑肌细胞收缩反应均低于对照组和治疗组,差异有统计学意义(均P<0.01);模型组CCK、NO及iNOSmRNA水平均高于对照组和治疗组,差异有统计学意义(P<0.01)。结论:马来酸曲美布汀可有效改善CP所致的胃肠动力障碍。
Objective: To explore the changes and mechanism of gastriointestinal
motility in rats with chronic pancreatitis, and the intervention effect of
trimebutine maleate on rat models. Methods: Thirty Wistar rats were randomly
divided into control, model and trimebutine maleate groups (n=10 for each group).
Rats of trimebutine maleate group were administered intragastrically with
trimebutine maleate (0.2 mL/kg,1/d) for two weeks. Rats of model and control group
were given same dose of saline in same way. The serum level of cholecystokinin
(CCK) was detected by enzyme-linked immunosorbent assay (ELISA). The level of
nitric oxide(NO) in cell cultures was quantified by the method of nitrate
reductase. The bowel movement coefficient, gastric and small intestinal circular
smooth muscle contraction ability and contractility of circular smooth muscle
cells were detected. The expression of inducible nitric oxide synthase (iNOS) mRNA
in gastrointestinal muscular was also detected. Results: The bowel movement
coefficient, gastrointestinal circular smooth muscle stripe contractility and
contractile response of the smooth muscle cells were significantly lower in model
group than those of control group and treatment group (P 〈 0.01). The serum levels
of CCK, NO and iNOS mRNA expression were significantly higher in model group than
those of control and treatment groups (P 〈 0.01). Conclusion: Trimebutine maleate
can effectively improve the gastrointestinal motility disorders caused by chronic
pancreatitis.
