摘要
用钙离子沉淀法制备猪肝细胞微粒体P450 进行了体外代谢隐丹参酮的研究。用Lowry 法测定肝微粒体蛋白含量平均为2 .460 ±0 .699 mg/ m L;Om ura 法测定猪肝微粒体P450 含量平均为0 .176 ±0 .053 n mol/ m g 。从P450加还原型辅酶Ⅱ的体外代谢系统中分离纯化了2 个主要代谢产物,经光谱分析鉴定为丹参酮ⅡA 与1羟基丹参酮ⅡA,并比较了隐丹参酮在猪体内外的代谢差异。
The pig liver microsomes P 450 were prepared by means of calcium ion precipitation in order to study in vitro metabolic transformation of Cryptotanshinone. The protein content of the liver microsomes was 2.460±0.66 mg/mL (n=5) by means of the Lowry's method; The average content of the liver microsomes P 450 was 0.176±0.053 nmol/mg ((n=5) by means of the Omura's method. The major metabolites were isolated by CC and preparative TLC, and identified as tanshinone Ⅱ A and 1 hydroxy tanshinone Ⅱ A by UV, EI MS and 1 HNMR spectral analysis. The difference of the metabolism profiles of Cryptotanshinone in vivo and in vitro was also compared.
