摘要
目的观察泄浊解毒方对溃疡性结肠炎大鼠肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、核因子-κB(nuclear factor-kappa B,NF-κB)、黏附分子-1(tntercellular adhesion molecule-1,ICAM-1)的影响并探讨其作用机制。方法 2,4,6-三硝基苯磺酸/乙醇混合溶液灌肠复制溃疡性结肠炎大鼠模型。实验设正常组、模型组、柳氮磺胺吡啶组、中药泄浊解毒方小、大剂量组;观察大鼠结肠病理形态学改变(疾病活动指数评分及黏膜损伤指数评分),采用放射免疫法测定TNF-α,运用免疫组织化学染色(SP法)检测NF-κB、ICAM-1表达。结果泄浊解毒方可明显降低大鼠结肠黏膜损伤指数评分,减少NF-κB、ICAM-1表达,降低TNF-α的含量。结论泄浊解毒方对大鼠溃疡性结肠炎有良好的治疗作用,其作用机制可能与减少NF-κB、ICAM-1表达,降低TNF-α的含量有关。
Objective To observe the effects of Xiezhuojiedu decoction on the expression of tumor necrosis factor - st ( TNF - st ), nuclear factor - kappa B ( NF - KB ), tntercellular adhesion molecule- 1 (ICAM -1 ) in ulcerative colitis rats, and study its mechanism. Methods 2,4,6 - trinitrobenzene sulfonic acid/ethanol solution was administrated by enema to make ulcerative colitis rats models. Five groups were set up in the experiment including normal control group, model control group, salazosulfamide group, Xiezhuojiedu decoction low -dose group, Xiezhuojiedu decoction high -dose group;The pathomorphological observation was carried out on the rats'colon disease activity index (DAI) scores and colon mucosal damage index (CMDI). TNF - st was determined by radioimmunoassay ; The expression of NF - KB and ICAM - 1 was determined by immunohistochemical staining. Results Xiezhuojiedu decoction can significantly decrease the CMDI scores, decrease the content of TNF- st, reduce the expression of NF - KB and ICAM - 1. Conclusion Xiezhuojiedu decoction has good therapeutic effect on ulcerative colitis in rats , its mechanism may be related to decreasing the content of TNF - st, and reducing the expression of NF - KB and ICAM - 1.
