摘要
目的:研究中枢神经系统两种大胶质细胞,星形胶质细胞及少突胶质细胞,对神经元轴突生长的影响。方法:取胚胎14 d 大鼠大脑皮质进行神经元与胶质细胞联合与分离培养,并培养嗜铬细胞瘤( Pheochrom ocytom a, P C12)细胞。分别取两种胶质细胞培养上清液、胶质细胞及用蒸馏水破碎的两种胶质细胞碎片加入培养的 P C12 细胞中。结果:两种培养细胞上清液、星形细胞及其碎片的加入对 P C12 细胞均无明显影响,但当少突胶质细胞及其细胞碎片加入到 P C12 细胞后在10 m in 内即出现明显的突起萎陷及回缩。另外,在联合培养中,少突胶质细胞附近的神经元不见明显的突起生长,且偶有神经元突起在遇到星形胶质细胞时不能继续延长,但无回缩现象。结论:少突胶质细胞的膜上存在着抑制神经元轴突生长的物质,这种物质不分泌到细胞外,并不依赖于该细胞的存活状态,而星形胶质细胞对神经元的突起生长具有机械性阻挡作用。
Objective: To investigate the effects of macroglial cells from rat CNS on axonal growth of neurons. Methods: Neuronal and macroglial cells from the cerebral cortex of rats on the 14th embryonic day were cultured. The oligodentrocytes were dissociated from astrocytes 9 days after the culture. The supernatants of two types of glia were collected on the 14th day of culture and added to the medium for culturing the PC12 cells. Observation was performed at the 10th min, 4th h and 12th h respectively. Results: The supernatants of the two types of glial cells had no significant effects on the neurites of PC12 cells. However, the oligodendrocyte and its debris had strong inhibitory effects on them at the 10th min, 4th h and 12th h as compared with the control group. The neurites of PC12 cells collapsed and became shorter as soon as the oligodendrocyte and its debris were added. In the primary culture, the neurons next to oligodendrocytes showed no neurites and outgrwoth. On the other hand, it was rarely found that the neurites of neurons could not extend but have no retract and collapse when the neurons reached the astrocytes. Conclusion: Some inhibiting substance may attach to the membrane of oligodendrocytes and it can inhibit the growth of neurites but can not be released into the supernatant. The astrocytes can only mechanically prevent the elongation of neurites.
