人参养荣汤对腺嘌呤致CRF营养不良模型大鼠下丘脑信号传导蛋白STAT3、PIAS3的影响 被引量:5

摘要 目的:探讨细胞因子信号传导蛋白STAT3及其抑制因子PIAS3在慢性肾衰竭营养不良大鼠中的表达情况,以及人参养荣汤对其的调节作用。方法:予0.5%腺嘌呤和4%酪蛋白混合饲料喂养制作CRF营养不良大鼠模型,将符合条件的随机分为模型组、人参养荣汤组和开同组,同时设正常组。各组分别予蒸馏水、人参养荣汤(含生药11.43 g/kg.d)、开同(1 g/kg.d)灌胃。治疗4周后检测白蛋白、血红蛋白的变化,Western免疫印迹法检测信号转导蛋白STAT3、PIAS3在下丘脑的表达。结果:人参养荣汤可增加白蛋白、血红蛋白水平,减低血浆瘦素水平,与正常组比较,模型组及人参养荣汤组STAT3表达均增强(P<0.001),人参养荣汤组STAT3表达显著低于模型组(P<0.001);模型组下丘脑PIAS3表达较正常组显著增强(P<0.001);人参养荣汤组PIAS3蛋白表达比模型组有显著增强(P<0.05)。结论:人参养荣汤通过上调CRF营养大鼠下丘脑PIAS3蛋白的表达水平,下调STAT3蛋白表达,从而进一步阻止瘦素受体JAK2/STAT3的信号传导,从而改善CRF营养不良状态。 Aim:To investigate the effects of Ginseng Yangrong soup on STAT3、PIAS3 in hypothalamus in malnutrition rats with chronic renal failure(CRF).Methods:Rats were fed with 0.5% adenine and 4% caseinum to make malnutrition in CRF models. Those consistents with malnutrition of CRF condition were randomized into CRF control group, Ginseng Yangrong soup group,Ketosteril group, and normal control group.after treatment for4 weeks as indicated, The blood parameters, like blood serum albumin(ALB), haemoglobin (Hb), The protein expressions of STAT3,PIAS3 were assessed respectively by immunoblotting.Results:the blood leptin in Ginseng Yangrong soup group decreased significantly while substantially blood serum ALB, Hb increased compared with the normal group rats ,the respective expression of STAT3 significiantly increased in CRF rats and Ginseng Yangrong soup group(P 0.001);After treatment with Ginseng Yangrong soup ,the expression of STAT3 in the CRF rats decreased significantly (P 0.001) compared with the normal group rats ,the respective expression of PIAS3 significiantly increased in CRF rats (P 0.001). compared with the CRF rats ,the respective expression of PIAS3 significiantly increased in Ginseng Yangrong soup rats (P 0.05) .Conclusion:PIAS3,STAT3 protein may be involved in the malnutrition associated withCRF .Upregulation of PIAS3 and reduction of STAT3 may be responsible for malnutrition effects of Ginseng Yangrong soup in CRF rats ,and the signal conduction of JAK2/STAT3 can be restrain.
