The influence of tensile stress on the differentiation of bone marrow stromal cells and the expression of mechano-growth factor

摘要 目的探讨张应力加载下骨髓基质细胞力生长因子(MGF)表达的影响及量效关系。方法骨髓基质细胞原代培养后加力不同时间经形态学观察,ELISA检测各组BMSC培养上清中ALP的浓度,Western blot检测加力后骨髓基质细胞MGF的表达。结果机械拉伸刺激下BMSC的形态学特性逐渐发生改变。加载0h、1h、3h、6h、12h、24h位移为1mm,频率为0.5Hz的机械张应力,均可增强BMSC的ALP活性,其中24h时ALP OD值最高(P<0.05)。Western Blot分析BMSC加力后MGF蛋白的表达,结果为细胞加力后MGF在1h开始有变化,MGF表达水平随加力作用时间延长而增高,组间比较差异有统计学意义(P<0.05)。结论 MGF可能通过刺激骨髓基质细胞ALP的分泌而促进其向成骨方向分化。 Objective To investigate the influence of tensile stress on the differentiation of bone marrow stromal cells(BMSC) and the expression of mechano-growth factor(MGF).Methods Primary culture of BMSC was observed morphologically in different time with tensile stress.The concentration of ALP in BMSC cultural supernatant was tested by ELISA,and the expression of MGF in BMSC after tensile stress by Western blot.Results Under the stimulus of mechanical drawing,BMSC gradually changed in morphological properties.Both loading with 0h,1h,3h,6h,12h and 24h,with 1mm displacements,and the tensile stress with frequency of 0.5Hz can enhance the expression of ALP in BMSC,with the highest ALP OD value at 24h(P0.05).Western blot analysis of the expression of MGF in BMSC showed that the cells began to change in MGF after 1h tensile stress,higher expression with longer period,with statistical significance between the groups(P0.05).Conclusion MGF may promote BMSC osteogenesis by stimulating the expression of ALP.
