摘要
目的 :探讨广西、海南和云南三地微小按蚊之间的基因差异。方法 :从三地单雌线蚊虫提取 DNA,然后分别用 Ava I、Bam HI、Pst I、Bgl I、Hinf I和 Hae 6种限制性核酸内切酶消化 ,进行琼脂电泳分析。结果 :三地微小按蚊的 Ava I、BamHI和 Pst I酶切区带有差异 ,Bgl I的酶切区带完全相同 ,而 Hinf I和 Hae 两种酶不显区带。结论 :三地微小按蚊有基因结构差异 ,Ava I、 Bam HI和 Pst I
Objective:To observe the variation in repetitive DNA of the An Minimus from Guangxi,Hainan and Yunnan Method:Total DNA was extracted from isofemale lines of the three strains and digested respectively with six restriction endonucleases (Ava I,Bam HI,Pst I,Bgl I,Hinf I and Hae Ⅲ).The DNA fragments were analysed by agarose gel electrophoresis coupled with ethidium bromide staining.The fluorescent DNA bands were visualized under a UV lamp and photographed Results:Banding patterns produced by Ava I,Bam HI and Pst I show some unique DNA fragments Bgl I digestion generally gave similar patterns The latter two enzymes gave vague patterns (results not shown) Conclusion:In view of the variation in repetitive DNA of different area strains of An minimus,Three enzymes(Ava I,Bam HI and Pst I) were capable of distinguishing among the three area strains
