摘要
采用PCR-DGGE技术对不同施肥处理毛竹根际微生物多样性特征进行研究。结果表明:采用改进的蛋白酶K-CTAB法提取的毛竹土壤DNA经PCR扩增的产物经DGGE检测后得到的电泳条带清晰且分离效果好,可以明显反映出毛竹根际微生物多样性的变化。根际微生物种群组成特征受施肥量、肥料种类影响很大,施肥能够增加毛竹根际微生物的多样性指数,在同等氮、磷养分情况下,钾肥的加入会对毛竹根际微生物种群组成产生很大的影响,并且根际微生物多样性有随钾肥施用量增加而升高的趋势。
A Polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE) method was used to determine the diversity of rhizosphere microorganisms of Phyllostachys edulis after different fertilizer application.Total community DNA was extracted and purified from soils by an improved protease K-CTAB method,and then amplified using eubacterial 16S rDNAs primers.Their PCR products were analyzed by DGGE to obtain bacterial community patterns.The electrophoresis lanes obtained by PCR-DGGE were legible and distinguishable,which distinctly indicated the change in rhizosphere microorganisms of Phyllostachys edulis.Results showed that rhizosphere microbial diversity could be affected significantly by fertilizer application.Rhizosphere microorganisms were significantly affected by the amount and type of fertilizers.Fertilizer application could increase Shannon-Wiener index of rhizosphere microorganisms.K fertilizer had a greater effect on microbes,and rhizosphere microbial diversity increased with increasing amount of K fertilizers.
