摘要
HIV疫苗研究的巨大障碍在于目前设计的包含Env蛋白或表位的疫苗在临床前及临床试验中难于诱导产生广谱中和抗体。近来的研究表明一些HIV-1感染者血清中存在的抗体能够中和多种病毒株,而新的表位作图技术对这些广谱中和血清表位特异性的深入分析将对广谱中和抗体靶向的病毒表位提供新的线索。通过广谱中和血清表位特异性研究获得的信息将促进新的广谱中和单克隆抗体的分离和潜在新表位的鉴定,为合理设计新的疫苗免疫原提供关键信息。
One obstacle to the development of an effective HIV-1 vaccine has been the difficulty in inducing broadly neutralizing antibodies with protective functions.However,recent studies show that antibodies in the sera of some HIV-1 infected individuals can neutralize diverse HIV-1 isolates.Detailed analyses of these sera with recently developed sophistical mapping methods can provide new insights into the viral epitopes targeted by broadly reactive NAbs.The new information emerging from these mapping efforts may be conducive to sharpening efforts to isolate new bnmAbs and moreover,may provide crucial information for the rational design of novel vaccine candidates.
