阿托伐他汀对HepG2细胞活性氧及转录因子Sp1mRNA表达的影响 被引量:1

摘要 目的探讨阿托伐他汀对HepG2细胞增殖、葡萄糖摄取、氧化应激和转录因子Sp1mRNA表达的影响及其分子机制。方法实验分为:对照组;阿伐他汀组。用MTT法检测对细胞增殖的影响;用微量化的葡萄糖氧化酶法检测细胞对葡萄糖的摄取作用;用流式细胞术检测细胞活性氧含量;用RT-PCR方法检测Sp1mRNA表达。结果阿托伐他汀作用48 h后对细胞增殖具有抑制作用(P<0.05);对细胞摄取利用葡萄糖具有促进作用(P<0.05);和对照组比较细胞活性氧含量显著减少(P<0.05);RT-PCR结果表明:转录因子Sp1mRNA表达和对照组比较明显增强(P<0.05)。结论阿托伐他汀对HepG2细胞增殖的抑制和改善葡萄糖利用作用可能与减轻氧化应激有关,可能与通过增强转录因子Sp1的表达导致相关基因的表达而产生的有利作用有关。
