摘要
目的通过观察新血府逐瘀汤对高血压大鼠内皮功能及心肌Ⅰ型、Ⅲ型胶原的影响,阐明活血化瘀中药干预高血压大鼠心肌纤维化的一种内在机制。方法建立腹主动脉缩窄致肾性高血压大鼠模型成功后,将40只大鼠分为五组,其中中药组、卡托普利组应用药物干预,模型组、假手术组、空白组不干预。10周后大鼠称重,腹主动脉抽血,测量左心室重量指数(LVWI)、血管性血友病因子(vWF)、内皮素(ET)、D-二聚体(D-D),免疫组织化学染色观察心肌Ⅲ型胶原和Ⅰ型胶原心肌组织胶原容积分数(CVF)、心肌血管周围胶原面积(PVCA)的改变。结果用药10周后,中药组大鼠血浆vWF、ET、D-D、血压以及LVMI与模型组比较,均明显下降(P<0.05或P<0.01)。卡托普利组和中药组的Ⅰ型胶原、Ⅲ型胶原CVF和PVCA较模型组下降(P<0.05)。结论新血府逐瘀汤可改善高血压大鼠血管内皮功能及高血压血栓前状态,减少心肌Ⅰ型、Ⅲ型胶原的形成,这可能是其防止高血压大鼠心肌纤维化的内在机制之一。
Objective To explain the internal mechanism on the invention for myocardial fibrosis treated with Chinese medicines for activating blood circulation and resolving stasis in the rats with hypertension through the observation of the effects of modified xuefu zhuyu decoction on endothelial function of myocardial collagen Ⅰ and Ⅲ in the rats with hypertension.Methods After the successful modeling the rats with renal hypertension induced by abdominal aorta stenosis,40 rats were randomized into 5 groups.Of them,the medicinal intervention was applied in a Chinese medicine group and a captoril group.No any intervention was performed in a model group,a sham-operation group and a blank group.10 weeks later,the body mass of rat was weighted and blood sample was collected from abdominal aorta.LVWI,vWF,ET and D-D were measured.ELISA was adopted to observe the changes in myocardial collagen Ⅲ and I.Results 10 weeks after medication,in Chinese medicine group,plasma vWF,ET and D-D,blood pressure and LVMI all decreased remarkably as compared with model group（P0.05 or P0.01）.In captoril group and Chinese medicine group,CVF and PVCA of collagen Ⅰ and Ⅲ decreased as compared with model group（P0.05）separately.Conclusion The modified xuefu zhuyu decoction improves endothelial function and the state before hypertensive thrombosis in the vessels of the rats with hypertension and reduces the formation of myocardial collagen Ⅰ and Ⅲ,which may be one of the internal mechanisms on the prevention of myocardial fibrosis in the rats with hypertension.
