摘要
目的观察威灵仙对人肝癌Bel7402增殖及Bcl-xl、Bcl-2蛋白表达的影响。方法将人肝癌Bel7402分为空白对照组、阳性对照组和威灵仙组,分别用普通培养液、氟尿嘧啶培养液、威灵仙培养液进行培养。采用四甲基偶氮唑盐比色法测定威灵仙对人肝癌细胞Bel7402的体外生长抑制率,并用免疫组化法检测三组细胞的Bcl-xl、Bcl-2蛋白表达量。结果威灵仙对人肝癌Bel7402细胞有显著增殖抑制作用,威灵仙组与空白对照组比较有统计学意义(P<0.01);威灵仙可抑制Bcl-xl、Bcl-2蛋白的表达,威灵仙组与空白对照组比较有统计学意义(P<0.01)。结论威灵仙可抑制Bel7402的生长,其机制可能与抑制Bcl-xl和Bcl-2蛋白的表达有关。
Objective To observe the effect of radix clematidis on proliferation of human hepatocellular carcinoma cell line Bel7402 and Bcl-2 and Bcl-xl protein expression.Methods The Bel7402 were divided into the untreated control group cultured with general culture fluid,the positive control group cultured with fluorouracil culture fluid and the radix clematidis group cultured with radix clematidis culture fluid,then adopt 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazoliumbromide to determine the ectogenic growth inhibition ratio of Bel7402 by radix clematidis,and detect protein expression of Bcl-xl,Bcl-2 with immunohisto chemical method.Results The studies demonstrate that radix clematidis inhibited proliferation of Bel7402 cells,which was a significant difference between radix clematidis group and normal control group(P〈0.01).And it could effectively decrease the level of Bcl-xl,Bcl-2 protein expression,which was a significant difference between radix clematidis group and normal control group(P〈0.01).Conclusion Radix clematidis can inhibit growth of Bel7402,the mechanism of which may be related to the inhibition of Bcl-xl and Bcl-2 protein expression.
