摘要
目的观察Snail蛋白及其mRNA在喉鳞癌组织中的表达及其与临床病理特征的关系,并探讨它在喉鳞癌发生发展和浸润转移中的作用及其与预后关系。方法应用原位分子杂交检测Snail mRNA和免疫组织化学S-P法检测Snail蛋白分别在60例喉鳞癌、20例癌旁组织中的表达。结果①Snail蛋白及其mRNA的表达与喉鳞癌的T分期、TNM分期、甲状软骨累及、淋巴结转移和远处转移有关(P均<0.05),但Snail mRNA的表达与病理分级无关(P>0.05),而Snail蛋白与病理分级有关(P<0.05),它们均与性别、年龄无关((P>0.05);②Snail蛋白与Snail mRNA在喉鳞癌组织中的表达明显正相关(r=0.362,P<0.05);③Snail蛋白阳性患者生存率明显低于阴性患者,其比较差异有统计学意义(P<0.05)。结论 Snail高表达可能是喉黏膜恶性转变以及喉鳞癌发生浸润转移的重要生物学标志,其高表达预示肿瘤有较高的侵袭性和不良预后。
Objective To investigate the expression of Snail protein and mRNA in laryngeal squamous cell carcinoma and its relationships with the clinico-pathological features,and to explore its roles in the carcinogenesis,progression and infiltration,metastasis of laryngeal squamous cell carcinoma and correlations with the prognosis.Methods In situ hybridization technique was used to detect the expression of Snail mRNA,and the immunohistochemical S-P methods to detect Snail protein in 60 cases of laryngeal squamous cell carcinoma and 20 cases of side cancer tissue.Results ①The expression of Snail protein and mRNA were correlated to T stage,TNM stage,thyroid cartilage involvement,lymph node metastasis and distant metastasis(all P0.05),but the expression of Snail mRNA was not correlated with pathologic stage(P0.05),and the expression of Snail protein was correlated with pathologic stage(P0.05),not with gender and age were irrelevant(all P0.05);②A positive correlation was found between Snail protein and mRNA in laryngeal squamous cell carcinoma(r=0.362,P0.05);③The survival rate of Snail protein positive patients was lower than that of negative patients,and its comparative differences was statistically significant(P0.05).Conclusion The overexpression of Snail might be important biological markers for malignant transformation and invasion and metastasis of laryngeal squamous cell carcinoma,and the up-regulation of the expression of Snail predicts more invasion and poor prognosis.
