摘要
目的探讨Toll样受体4(Toll-like receptor 4,TLR4)及p-Akt在哮喘发病中的作用及小青龙汤对气道平滑肌迁移的影响。方法 SPF级♂大鼠40只随机分为:正常对照组(A)、哮喘4周组(B)、哮喘8周组(C)、小青龙汤4周组(D)和小青龙汤8周组(E),每组8只。以卵清白蛋白(OVA)致敏与激发建立哮喘大鼠气道重塑模型。测定气道阻力;观察气道壁嗜酸性粒细胞侵润情况;图像分析软件测定支气管管壁面积、平滑肌面积和平滑肌至上皮下基底膜距离;免疫组织化学方法检测TLR4、p-Akt蛋白表达。结果 B组、C组、D组和E组大鼠的WAt/Pi、WAm/Pi较正常对照组均增加(P<0.01),间距/Pi减小(P<0.01);B组和C组大鼠的WAt/Pi高于E组,间距/Pi则较两组明显缩短(P<0.05);TLR4在B组和C组气道平滑肌上呈阳性或强阳性表达,表达量高于A组(P<0.01),C组亦高于B组(P<0.05),E组的表达量亦低于C组和D组(P<0.01)。p-Akt在B组、C组及D组的表达均高于A组(P<0.01),D组亦高于B组(P<0.05),E组低于B组和D组,与D组相比亦降低(P<0.05)。结论 TLR4和p-Akt参与哮喘气道平滑肌的迁移,小青龙汤可能通过抑制TLR4和p-Akt的表达在哮喘气道重塑方面发挥一定的作用。
Aim To explore the effect mechanism of TLR4 and p-Akt on the airway smooth muscle migration and the impact of Xiaoqinglong decoction on the migration in asthmatic rats.Methods 40 rats were divided into five groups including a control group(group A),4 weeks of asthmatic model group(group B),8 weeks of asthmatic model group(group C),4 weeks of Xiaoqinglong decoction group(group D) and 8 weeks of Xiaoqinglong decoction group(group E),with 8 rats in each group.The chronic asthmatic model was established by repeated inhalation of ovalbumin.The airway resistance was detected;the airway eosinophilic inflammation of airway wall was observed;the airway wall area,the bronchial smooth muscle area and the distance of smooth muscle to basement membrane of subcutaneous were measured by image analysis system;the expression of TLR4 and p-Akt protein was determined by immunohistochemical staining.Results The WAt /Pi and WAm/Pi in B group,C group,D group and E group were thicker than those in A group(P〈0.01),while the distance/Pi was shorter(P〈0.01).The WAt /Pi in B group and C group was thicker than that in E group,while the distance/Pi was shorter(P〈0.05);the content of TLR4 in airway smooth muscle of B group and C group was more than that of A group(P〈0.01) while C group was more than B group(P〈0.05).The content of TLR4 in airway smooth muscle of E group was less than that of C group and D group(P〈0.01).The content of p-Akt in airway smooth muscle of B group,C group and D group was more than that of A group(P〈0.01) while C group was more than B group(P〈0.05).The content of p-Akt in airway smooth muscle of E group was less than that of B group and C group and less than that of D group(P〈0.05).Conclusion TLR4 and p-Akt participate in the migration of asthmatic airway smooth muscle and through inhibiting TLR4 and p-Akt expression,Xiaoqinglong decoction may play a role in the airway remodeling of asthma.
