摘要
为观察不同浓度紫杉醇诱导犬乳腺肿瘤细胞凋亡的形态学变化,以体外培养犬乳腺肿瘤系CHMm株为模型,分别用0.01、0.1、1μmol/L紫杉醇与犬乳腺肿瘤细胞作用48h后,分别用普通光学显微镜观察细胞形态变化、BrdU标记法载玻片观察细胞增殖状态、AO/EB双荧光染色荧光显微镜及电镜观察犬乳腺肿瘤细胞的凋亡情况。结果显示,不同浓度紫杉醇作用后,CHMm细胞生长受到不同程度的抑制,并随浓度的增加,出现典型的凋亡形态。结果表明,紫杉醇能抑制CHMm生长,引起细胞损伤,诱导细胞凋亡,并具有浓度依赖性。
To investigate Taxol-induced apoptosis in canine mammary tumours,canine breast cancer cell line CHMm was treated with Taxol at 0.01,0.1 and 1 μmol/L for 48 hour,respectively.Morphological changes were monitored using the light microscope.Apoptosis was quantified using the AO/EB double staining,followed by imaging and counting apoptotic cells with a fluorescent microscope.Cell proliferation was measured by the BrdU methodology.In result,Taxol inhibited the proliferation of canine breast cancer cells and induced apoptosis,with the appearance of typical morphological features of apoptosis,in canine breast cancer cells in a dose-dependent manner.In conclusion,Taxol inhibited cell proliferation,and dose-depen-dently induced cellular injuries and apoptotic cell death.
