摘要
目的:探讨胰岛素在高糖环境下牙周膜细胞凋亡过程中的调节作用。方法:本实验采用含有不同葡萄糖浓度的培养基培养人牙周膜细胞,并采用胰岛素作治疗对照,作用24小时后,立即使用流式细胞仪对各组凋亡细胞进行计数,最后进行统计学分析。结果:葡萄糖浓度增至15mmol/L时,牙周膜细胞凋亡比例无显著增加(3.47±0.78 VS 3.03±0.40,P>0.05),当葡萄糖浓度增至25mmol/L和35mmol/L时,牙周膜细胞凋亡比例显著增加(5.07±0.611 VS 3.03±0.40,P<0.01;7.13±0.72 VS 3.03±0.40,P<0.01),加入胰岛素后,牙周膜细胞凋亡比例显著下降(7.13±0.72 VS 5.23±0.85,P<0.01),但仍然高于正常糖浓度组(5.23±0.85 VS3.03±0.40,P<0.01)。结论:胰岛素能够部分抑制高糖环境诱发的牙周膜细胞凋亡。
Objective: To investigate the regulation effect of insulin in human periodontal ligament fibroblasts apoptosis caused by high glucose.Methods: Human periodontal ligament cells were cultured in medium with different concentration of glucose for 24h,insulin was used as treatment control.Then we used flow cytometer to count the percentage of apoptotic cells of each team immediately.Then we performed statistical analysis.Results: When glucose concentration increased to 15mmol/L,apoptotic ratio of periodontal ligament fibroblasts did not change significantly(3.47±0.78 VS 3.03±0.40,P〉0.05).When glucose concentration increased to 25mmol/L and 35mmol/L,apoptotic ratio of periodontal ligament fibroblasts increased significantly(5.07±0.611 VS 3.03±0.40,P〈0.01,7.13±0.72 VS 3.03±0.40,P 〈0.01).After adding insulin,periodontal ligament fibroblasts apoptotic ratio decreased significantly(7.13±0.72 VS 5.23±0.85,P 0.01),but still higher than the team of normal glucose concentration(5.23±0.85 VS 3.03±0.40,P 0.01).Conclusion: Insulin could partly inhibit human periodontal ligament fibroblasts apoptosis caused by high glucose.
