摘要
目的:探讨体外细胞培养,观察核转录因子κB通路特异性抑制剂吡咯烷二硫代氨基甲酸酯(PDTC)对膀胱癌EJ细胞凋亡抑制蛋白Livin、Survivin mRNA表达水平的影响。方法:采用Real-time PCR法检测以不同浓度不同时间的PDTC处理的膀胱癌EJ细胞的凋亡抑制蛋白Livin、Survivin mRNA表达水平情况。结果:PDTC可降低膀胱癌EJ细胞凋亡抑制因子Livin、Survivin mRNA表达水平,呈剂量-时间依赖性,且呈负相关(P<0.05)。结论:PDTC可使膀胱癌EJ细胞凋亡抑制蛋白Livin、Survivin mRNA表达水平减少,其机制可能与NF-κB通路被抑制相关。
Objective: Cell culture in vitro,to investigate the effects of PDTC,the selective inhibitor of NF-κB kinase,on mRNA expression of apoptosis inhibited protein Livin and Survivin on human bladder carcinoma cell EJ.Methods: The method of Real-time PCR for detecting the mRNA expressions of Livin and Survivin was used.Results: When PDTC was given,the mRNA expression of Livin and Survivin were down-regulated in a dose-and time-dependent manner(P0.05).Conclusion: PDTC can down-regulate mRNA expression of apoptosis inhibitory protein Livin and Survivin on bladder carcinoma cell EJ,which might be performed by inhibiting NF-κB signal path by PDTC.
