摘要
目的探讨干扰素/维甲酸联合应用诱导的细胞凋亡相关基因19(GRIM-19)和信号转到子与转录激活子3(STAT3)蛋白在肝细胞癌中的表达、相互关系及在肝细胞癌发生发展中的意义。方法应用免疫组化方法检测GRIM-19和STAT3在31例肝细胞癌组织和癌周肝组织、20例肝硬化组织及10例正常肝组织的表达水平。结果肝硬化组织中GRIM-19蛋白表达强度显著高于肝细胞癌组织和正常肝组织(P〈0.05),肝细胞癌组织中GRIM-19蛋白表达强度与癌周肝组织差异无统计学意义,肝硬化组织中GRIM-19蛋白表达强度与癌周肝组织差异也无统计学意义;肝细胞癌组织STAT3蛋白阳性表达高于癌周肝组织、肝硬化组织和正常肝组织(P〈0.05)。肝细胞癌组织中GRIM-19的表达与肿瘤TNM分期相关,GRIM-19在Ⅰ~Ⅱ期表达高于Ⅲ~Ⅳ期(P〈0.05)。肝细胞癌组织中GRIM-19和STAT3表达具有显著负相关(r=-0.468,P〈0.05)。结论肝细胞癌组织中存在STAT3的持续高表达和GRIM-19的低表达共存现象,GRIM-19作为一个新的细胞凋亡促进基因,可能参与了肝硬化、肝细胞癌的发生过程,可能成为一种新的肿瘤标记物,并可用于肝细胞癌的早期筛选。
Objective To investigate the expression of gene associated with retinoid-IFN-induced mortality-19(GRIM-19)and signal transducer and activator of transcription3(STAT3)in hepatocellular carcinoma(HCC),as well as their significance in the occurrence and development HCC.Methods The expression of GRIM-19 and STAT3 proteins were examined by immunohistochemistry in 31 tissue specimens from HCC and their para-carcinoma tissues,20 cases from liver cirrhosis tissues,and 10 cases from normal liver tissues.Results The positive rate of GRIM-19 expression was significantly higher in the liver cirrhosis tissues than that in the HCC tissues and normal liver tissues(P〈0.05),but GRIM-19 expression was not significant between the HCC tissues and para-carcinoma tissues,its expression was also not significant between the liver cirrhosis tissues and para-carcinoma tissues.While the positive rate of STAT3 expression was markedly higher in HCC than that in para-carcinoma tissues,liver cirrhosis tissues and normal liver tissues(P〈0.05).Expression of GRIM-19 was related to TNM stage of HCC(P〈0.05),the relative amount of GRIM-19 proteins expression of stage Ⅰ~Ⅱ were significantly higher than those of stage Ⅲ~Ⅳ.GRIM-19 expression in HCC was negatively correlated to STAT3 expression(r=-0.468,P〈0.05).Conclusion The high expression of STAT3 and the low expression of GRIM-19 co-exist in HCC,GRIM-19 may participate in the tumorigenesis and development of HCC and hepatic cirrhosis,and GRIM-19 may be as a new tumor marker to HCC early screening.
