摘要
目的:观察补肾健脾活血法对超排卵小鼠子宫内膜白血病抑制因子(LIF)表达的影响。方法:雌性小鼠予阿拉瑞林/孕马血清促性腺激素/人绒毛膜促性腺激素构建超排卵小鼠模型,并随机分成实验组(中药组)、阳性对照组(阿司匹林组)、阴性对照组,分别给予补肾健脾活血中药、阿司匹林、0.9%氯化钠注射液进行干预。在小鼠种植窗期取子宫中段组织,用逆转录-PCR(RT-PCR)法检测小鼠子宫内膜组织中LIF mRNA的表达水平。结果:中药组与阿司匹林组超排卵小鼠子宫内膜LIF mRNA的表达均明显高于阴性对照组,差异有显著统计学意义(P<0.01);中药组LIF mRNA的表达高于阿司匹林组,差异有统计学意义(P<0.05)。结论:补肾健脾活血法可以提高超排卵状态下子宫内膜LIF mRNA的表达水平,改善超排卵状态下子宫内膜的容受性,效果优于阿司匹林。
Objective:To observe the effect of strengthening spleen and kidney and activating blood method on the expression of leukemia inhibiting factor(LIF) in endometrium of controlled ovarian hyperstimulation(COH) mice at implantation window stage.Methods:The female mice were used to establish mouse models of COH by treated with Alarelin,pregnant mare serum gonadotrophin and human chorionic gonadotropin,then the mice were divided into experimental group(traditional Chinese medicine group,the mice were treated with strengthening spleen and kidney and activating blood method),positive control group(aspirin group,the mice were treated with aspirin) and negative control group(the mice were treated with 0.9% sodium chloride injection).The middle tissues of cervix at implantation window stage were obtained,RT-PCR was used to detect the expression level of LIF mRNA in endometrium of mice.Results:The expression levels of LIF mRNA in traditional Chinese medicine group and aspirin group were significantly higher than that in negative control group(P0.01);the expression level of LIF mRNA in traditional Chinese medicine group was significantly higher than that in aspirin group(P0.05).Conclusion:Strengthening spleen and kidney and activating blood method can increase the expression level of LIF mRNA in endometrium and improve the receptivity of endometrium of COH mice,and its effect is superior to that of aspirin.
