摘要
目的观察2型糖尿病小鼠胰腺中细胞核因子(nuclear factor kappa B,NFκBp65)的表达及意义。方法引进日本C57b1/Ksj-db/+m表型正常隐性基因小鼠,近亲(兄妹)交配,其纯合子1/4后代-db/db小鼠表现为肥胖和高血糖,即2型糖尿病模型。用免疫组化方法,观察各年龄段小鼠胰腺中NFκBp65的表达情况。结果 NFκBp65的阳性反应产物位于胰岛细胞的胞浆内随着月龄的增加表达增强,糖尿病组的表达明显高于正常组,同月龄糖尿病组阳性细胞平均光密度值与正常组比较,差异有统计学意义(P<0.05),糖尿病组间阳性细胞平均光密度值比较差异有统计学意义(P<0.05)。正常组间阳性细胞平均光密度值比较差异无统计学意义(P>0.05)。结论 2型糖尿病小鼠胰腺中NFκBp65的表达增强可能是糖尿病的发病机制之一。
Objective To investigate the expression and significance of NFκBp65 in the pancreas of mice with type 2 diabetes mellitus(T2DM).Methods The experimental animals were C57bl/Ksj-db/+m mutant diabetic mice imported from Japan,and consanguinity copulation was performed strictly.The homozygote 1/4 offsprings were db/db mice who displayed obesity and high blood glucose,i.e.T2DM animal models.Immunohistochemistry technique was used to detect the expression of NFκBp65 in the pancreas of mice models.Results The NF κ Bp65 positive reaction products were located in the pancreatic islet cells,and its expression was increased with the increase of month's age.The expression of NF κ Bp65 positive reaction products in diabetes group was significantly higher than that in control group,and there was a significant difference in the average optical density(OD) value at the same month's age between diabetes group and control group(P0.05),and there were significant differences among diabetes groups(P0.05),however,there was no significant difference among control groups(P0.05).Conclusion The enhancement of the expression of NF κ Bp65 in the pancreatic islet of mice with type 2 diabetes mellitus may be one of the pathogeneses of diabetes mellitus.
