摘要
目的观察湿热痹片对佐剂性关节炎(Adjuvant Arthritis,AA)大鼠关节滑膜组织中基质金属蛋白酶-1(MMP-1)及基质金属蛋白酶-3(MMP-3)表达的影响。方法以Mtb免疫SD大鼠诱导类风湿关节炎(Rheumatoid Arthritis,RA),灌胃给予相应药物,连续28天。采用核酸原位杂交法检测MMP-1和MMP-3在大鼠关节滑膜组织中的表达,经图象分析以平均光密度(MIOD)做半定量测量。结果湿热痹片高剂量组能显著降低AA大鼠滑膜组织中MMP-1、MMP-3的MIOD(P<0.01),湿热痹片低剂量组对MMP-1也有降低作用(P<0.05)。结论湿热痹片对AA大鼠滑膜组织中MMP-1、MMP-3的高表达有显著的改善作用。
Objective To investigate the effects of Shi-Re-Bi tablet on the expression of matrix metalloproteinase-1(MMP-1) and matrix metalloproteinase-3(MMP-3) of synovium tissue in adjuvant arthritis(AA) rats.Methods The model of AA in SD rats were made by immunized s.c.at the base of the tail with 200 μl of Mtb in mineral oil and treated with Shi-Re-Bi tablet and Leflunomide tablet.28 days later,the expression of MMP-1 mRNA and MMP-3 mRNA in the synovium tissue were detected by using situ hybridization.Results Compared with the model group,the high-dose of Shi-Re-Bi tablet could significantly decrease the MIOD of MMP-1 and MMP-3 of synovium tissue in AA rats(P 〈0.01),the low-dose could reduce the MIOD of MMP-1(P〈0.05).Conclusion Shi-Re-Bi tablet has a positive effects,it can decrease the expression of MMP-1 and MMP-3 of synovium tissue in AA rats.
