摘要
[目的]研究蚯蚓体内植酸酶的来源,为其开发利用提供借鉴。[方法]将干蚯蚓与0.9%NaCl溶液按照1∶4的比例在50℃条件下反应8 h,获得蚯蚓自溶液,再稀释得到10-1、10-2、10-33个浓度梯度溶液,涂布在改良的细菌培养基、合成培养基、改良的查氏培养基及含有植酸钙的琼脂平板上,在不同温度下培养。[结果]在改良的查氏培养基上,28℃培养5 d,出现了透明菌圈,且该透明菌圈随着培养时间的延长而逐渐扩大;在改良的细菌培养基、合成培养基及琼脂平板上均无透明菌圈出现。[结论]蚯蚓体内植酸酶来自于蚯蚓体内的真菌,而非细菌产生或蚯蚓自身产生。
[Objective] The aim was to study the source of phytase in the earthworms,to provide reference for development and utilization.[Method] The dry earthworm was mixed with 0.9% NaCl solution according to the ratio of 1∶4 and acted at 50 ℃for 8 h,and then the earthworm solution was diluted by 10-1,10-2,10-3.The earthworm solutions were coated respectively in the modified medium for bacteria,synthetic medium,modified Czapek and the agar plates which contains phytate-calcium.These plates were incubated at different temperature.[Result] The experimental results showed that the bacteria and fungi transparent circle was appeared in the modified Czapek medium which was cultured 5 d at 28 ℃.And that expanded gradually with the bacteria growing.There didn't appear clear circle in the modified medium for bacteria,synthetic medium and agar plates.[Conclusion] The phytase in the earthworm comes from the fungi,rather than bacteria and earthworm itself.
