摘要
目的:通过观察杞菊合剂对自发性高血压大鼠(SHR)血压、左室重量与体重比(LVW/BW)、心肌组织原癌基因N-ras及c-myc mRNA表达的影响,探讨该方剂对高血压左室肥厚的作用及其机理。方法:SHR 32只,随机分成杞菊合剂组(A组,8只)、苯那普利组(B组,8只)、杞菊合剂加苯那普利组(C组,8只)、模型组(D组,8只),再取8只正常SD大鼠作为正常对照组(E组,8只),共五组。各治疗组给予相应药物剂量灌胃,正常组与模型组同时给予等量的蒸馏水灌胃,每日1次,治疗12周,每周测量血压,实验结束时收集标本,检测左室重量与体重比(LVW/BW)、心肌组织原癌基因N-ras及c-myc mRNA的表达。结果:杞菊合剂可降低SHR的血压、左室重量、心肌组织N-ras c-myc mRNA的表达(P<0.05)。以杞菊合剂加苯那普利疗效最佳,杞菊合剂在降压方面不及苯那普利,二者对左室重量及N-ras、c-myc mRNA表达的影响与苯那普利无显著性差异(P﹥0.05)。结论:杞菊合剂可有效逆转高血压左室肥厚;杞菊合剂逆转高血压左室肥厚的机制可能与其降低血压,抑制原癌基因N-ras、c-myc表达有关;杞菊合剂与苯那普利联合应用其逆转高血压左室肥厚的作用更为显著。
Objective:Threugh observing the effect of Qi Ju mixture on the blood pressure(BP) ,left ventricular weight (LVW) to body weight(BW) ration(LVW/BW) and the expression of c - myc and N - ras mRNA in cardlomyocytes tissue in spontanenous hypertention rats ( SHR), we protected the mechanism of Qi Ju mixture for p [ reventing left ventricnlar hypertrophy by the experiment. Methods:Thirty two SHR were randomly divided into four groups:Qi Ju mixture group "A group" ( 8 rats), benazepril group "B group" ( 8 rats ), Qi Ju mixture and benazepril group "C group" ( 8 rats ), model group "D group" (8 rats) ;and other eight SD rats was the normal contrast group "E group" . The respective dose and drugs were given to the treated groups , and the same dose of distilled water was given to the normal and model groups by stomach rearing once per day for 12 weeks. During the experiment, the BP was measured at every week. After therapy ,the left ventrieular weight to body weight ration(LVW/BW) and the expression of c -myc and N -ms mRNA in cardiomyo- cries tissue of each group rats were tested. Results : Qi Ju mixture could low erblood pressure, the left ventricular weight, the expression of c -myc and N -ras mRNA in cardiomyocytes tissue of SHR( P 〈 0.05 ). The function of group C was the best in the three treated groups ( P 〈 0.05 ). Although Qi Ju mixture was weaker than benazepril in lowering the blood pressure (P 〈 0.05 ) , there were no significant difference in lowering blood pressure, reducing the left ventrlenlar weight, the expression of e - mye and N - ras mRNA in cardiomyoeytes tissue ( P 〉 0.05 ). Conclusion: ( 1 ) Qi Ju mixture could prevent hypertensive LVH. (2) Qi Ju mixture may be play important roles in preventing the LVH such as lowering blood pressure and down - regulating the expression of e - mye and N - ras. ( 3 ) The effect of Qi Ju mixture and benazeprll on preventing LVH was more better than Qi Ju mixtu
