摘要
为了研究柠檬酸对小鼠肝脏组织氧化应激的影响,将40只健康昆明品系小白鼠随机分为对照组(生理盐水),柠檬酸高(480 mg/kg)、中(240 mg/kg)、低(120 mg/kg)剂量组。各组小鼠每周腹腔注射相应试剂0.2 mL,连续2周,第2次注射7 d后取肝组织制作匀浆检测总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,同时制作石蜡切片,观察各组小鼠肝脏组织的形态变化。结果表明,与对照组相比,高剂量柠檬酸组小鼠肝脏组织T-SOD和GSH-Px活性显著降低,各剂量组MDA含量差异不显著。显微镜观察可知,柠檬酸组小鼠出现肝细胞索排列紊乱,中央静脉大量淤血,周围淋巴细胞和中性粒细胞浸润等一系列病理变化。
To study effect of ctric acid on oxidative stress of liver in mice,40 health Kunming mice were randomly divided into control group(0.9% saline),high dose group(480 mg/kg),middle dose group(240 mg/kg) and low dose group(120 mg/kg).All the mice were received an injection of 0.2 mL corresponding reagent each week for 2 weeks.After 7 days since the second injection,T-SOD,GSH-Px activities,MDA content were detected with liver tissue of mice,and morphologic change were observed after making paraffin section.Compared with control group,the activities of T-SOD,GSH-Px in high dose group were significant degraded,and MDA content in every group showed no significant difference.Microscopical examination of the liver showed that a series of pathological change were observed in mice of ctric acid group,such as indiscriminate liver cell cord,blood clot in central veins,lymphocyte and neutrophil infiltrating.
