摘要
目的观察大鼠严重烫伤延迟复苏后肺组织缺氧诱导因子-1α(HIF-1α)的表达变化,探讨其与肺组织损伤的相关性。方法雄性SD大鼠120只,随机分为延迟复苏组(DF,n=60)、即时复苏组(IF,n=50)和正常对照组(NC,n=10),建立Ⅲ度烫伤面积30%TBSA的模型,分别于伤后1、6、12、24、48和72h取材,采用组织病理学、组织芯片技术、免疫组织化学染色与图象分析技术,观察肺组织病理改变与HIF-1α的表达。结果 DF组伤后6~24h肺组织病理损伤严重,IF组相对较轻。HIF-1α的阳性表达位于肺泡上皮细胞胞核,DF组表达明显强于IF组,而NC组表达阴性。HIF-1α表达的灰度值定量分析结果显示:DF组与IF组在伤后72h各时相点表达均升高,与NC组比较差异有统计学意义(P<0.05);DF组在各时相点HIF-1α表达均高于相对应的IF组各时相点,差异有统计学意义(P<0.05)。结论严重烫伤延迟复苏后大鼠肺组织HIF-1α的高表达与肺组织的损伤的程度是一致的,推测其参与了严重烫伤延迟复苏后大鼠肺组织的损伤。
Objective This study aimed to observe the changes of hypoxia-inducible transcription factor- 1α (HIF-1α)expression after severe burn with delayed fluid resuscitation at different time-points, and explore the correlation between HIF-1α expression and lung tissue injury. Methods A total of 120 male SD rats were employed as the experimental models (30% TBSA, full-thickness burn wound), and they were randomly divided into 3 groups: delayed fluid resuscitation group (DF), immediate fluid resuscitation group (IF) and normal control group (NC). The lung samples were harvested at 1,6, 12, 24, 48 and 72 h after burn respectively. Pathological changes and HIF-1α expression were detected by using histopathology, tissue microarry, immunohistochemistry and image analysis techniques. Results At posthurn 6-24 h, the pathological changes were more severe in DF group than those in IF group. HIF-1α were much higher in DF group and IF group than those in NC group (P 〈 0.05), also higher in DF than in IF on every phase points posthurn 72 h (P 〈0.05). The increase of HIF-1α were observed in nucleus and cytoplasmic. Conclusions After severe burn with delayed fluid resuscitation in rats, the higher expression of HIF-1α and the extent of lung injury were consistent. Therefore, we infer that HIF-1α participates in lung injury in rats after severe burns with delayed fluid resuscitation.
