多囊卵巢综合征大鼠颗粒细胞自噬率的相关研究 被引量:8

摘要 目的了解多囊卵巢综合征(PCOS)大鼠及正常大鼠颗粒细胞自噬的差异,从而探索PCOS发生的相关机制。方法 20只SD大鼠随机分为2组,10只建立来曲唑诱导PCOS大鼠动物模型(模型组),10只作为对照组,21d后抽取腹主动脉血,进行血清学指标测定,提取模型组与对照组大鼠卵巢颗粒细胞,MDC荧光染色及流式细胞术测定大鼠颗粒细胞自噬率,并比较其差异。结果模型组大鼠体质量高于对照组,血清睾酮、卵泡刺激素水平均高于对照组,差异有统计学意义(P<0.05,P<0.01),模型组大鼠卵巢颗粒细胞自噬率(18.6%)明显高于对照组(3.6%),差异有统计学意义(P<0.05)。结论多囊卵巢综合征大鼠卵巢颗粒细胞的减少与颗粒细胞的自噬相关。 Objective To observe the differences between the polycystic ovary syndrome(PCOS) and normal rat granulosa cells 'autophagy rate,in order to investigate the mechanism of PCOS.Methods Use letrozole to induce the PCOS in rat model(n = 10),and 10 rats were enrolled as control.After 21 days,extract the ovarian granulosa cells in model and normal rats,use MDC staining and flow cytometry to determine the rate of rat granulosa cells autophagy,then compare the differences between the two groups.Results The weight of rat in model group was higher than control group,the level of hormones in the model group was higher that of control group(P0.05,P0.01 ).Model group rat granulosa cells autophagy rate (18.6%) was significantly higher than that of normal group(3.6%,P0.05 ).Conclusion The decrease of ovarian granulosa cells in PCOS rat were related with the autophagy of granulosa cells.
