摘要
目的:观察"康宁"胶囊(现已改名为丹芪活肝胶囊)对四氯化碳(CCl4)致小鼠急性肝损伤的的保护作用。方法:取KM小鼠72只,随机分为正常对照组、模型组、联苯双酯组、丹芪活肝胶囊高剂量组、中剂量组、低剂量组6组,每组12只。小鼠分别灌胃给药,1次/d,连续5 d,末次给药24 h后,除正常对照组外,其余各组小鼠均腹腔注射0.05 g/kg的CCl4花生油溶液,24 h后采血,检测血清ALT、AST活性及观察肝组织病理学变化。结果:丹芪活肝胶囊能降低CCl4所致小鼠肝损伤中血清ALT或AST活性(P<0.01~0.05)。丹芪活肝胶囊高、中、低剂量组均能明显改善小鼠肝组织损伤的程度。结论:丹芪活肝胶囊对CCl4诱导的小鼠肝损伤有保护作用,其作用机制可能与保护肝细胞膜、减轻炎性反应有关,具体机制还需进一步实验研究。
Objective:To observe the protective effect of Danqi Huogan Capsule(DHC) on liver injury in mice induced by carbon tetrachloride(CCl4).Methods:72 KM mice were randomly divided into six groups:normal control group,model group,bifendate group,high dosage group of DHC,medium dosage group of DHC,and low dosage group of DHC.All mice were administered with DHC by intragastric administration(ig),one time per day for five consecutive days,and 24 hours following the last dosage of administration,the mice in groups except the normal control group were administered with CCl4(0.05 g/kg) by ip.Blood samples were collected in 24 hours following the ip injection and the levels of ALT,AST activity content in serum were measured,and the pathological change of liver tissue were observed.Results:DHC has lowered the levels of ALT and AST activity significantly in the serum of the mice with liver damage induced by CCl4(P0.05,P0.01).In addition,the pathological sections showed that the liver injury in both high and medium dosage DHC groups has been significantly improved.Conclusions:DHC can protect against liver injury induced by CCl4 in mice.The mechanism may be related to protection of liver cell membrane and reduction of the inflammatory response.
