摘要
从云南昆明、元阳、玉溪、昭通地区采集豆豉样品26个,通过琼脂-纤维蛋白平板试验,SDS-fibrin-PAGE蛋白电泳结合Zymography活性染色对相应酶谱进行分析;同时采用茚三酮法测定其豆豉纤溶酶活性,筛选得到5株豆豉纤溶酶活性较高的菌株,其中尤以ZT-13菌株纤溶酶活性达到97.09 U/mL,是工业化生产豆豉纤溶酶的潜在优良菌株。
26 Douchi samples were collected from Kunming,Yuanyang,Yuxi,Zhaotong region and Yunnan province.By agar-fibrin plate test,SDS-fibrin-PAGE protein electrophoresis combining zymography staining to analyze the pattern of secreted enzymes.Meanwhile,the fibrinolytic activity was determined by Ninhydrin method.There were 5 strains screened with high-yield fibrinolytic activity.Especially,the ZT-13 strain of fibrinolytic activity was high and reached 97.09 U/mL.It is a potential strain for producing industrial fibrinolytic enzyme.
