摘要
目的:观察几丁质作为植骨材料的生物相容性。方法:通过体外实验与体内实验相结合的方法研究。结果:几丁质对人成骨细胞具有良好的相容性,不影响人成骨细胞的增殖及ALP活性,人成骨细胞可以在几丁质膜表面较好地贴附和伸展,但增殖较慢。培养40d时可以形成矿化结节。几丁质植入肌肉中,周围组织无明显炎症反应,未发现组织变性和坏死,并可完全降解。结论:几丁质具有较好的骨性生物相容性,但仍需提高其表面活性。
Objective: To investigate the biocompatibility of chitin as bone substitute material. Methods: In vitro and in vivo model was employed for the study. Results: Extraction of chitin had no poisonous effect on the proliferation and ALP activity of human osteoblasts,and cells adhered and expanded well on the surface of chitin,but proliferated slowly;mineralized nodule was found 40 days after cells had been seeded on the membrane.In the in vivo test it was observed that chitin had no toxicity on surrounding tissue,and soft tissue could soon grow into its pores;20 mg chitin was completely degraded 6 weeks after implanted subcutaneously into mouse back and 10 weeks into mouse leg muscular porch. Conclusion: Chitin may be suitable for bone substitute material,but its surface shoud be modified to meet the requirments for bone inductive activity.
