摘要
研究不同采收期栽培与野生无梗五加叶中总黄酮及多糖含量的变化,为更好地开发利用无梗五加资源提供理论依据。采用紫外分光光度法,以芦丁作为对照品,在500nm波长下测定不同采收期栽培与野生无梗五加叶中总黄酮的含量;采用蒽酮-硫酸比色法,以葡萄糖作为对照品,在620nm波长下测定不同采收期栽培与野生无梗五加叶中可溶性多糖和粗多糖的含量;采用SPSS16.0统计分析软件对试验结果进行统计分析。在0.05mg^0.5mg范围内,芦丁质量(mg)与吸光度A值线性关系良好,R2=0.9996;在0.0208mg^0.104mg范围内,葡萄糖质量(mg)与吸光度A值线性关系良好,R2=0.9996;结论:栽培无梗五加叶中黄酮及多糖含量均略高于野生无梗五加叶但含量差异并不显著。同时从方法学考察可知,本方法简便、快速、可靠、易行、重现性好,适合无梗五加叶总黄酮及多糖的含量测定。
To study the content of total flavonoid and polysaccharides in leaves of Acanthopanax sessiliflorus produced in different growth stages.To provide a theoretical basis in order to better use the resource of Acanthopanax sessiliflorus.The content of total flavonoid and polysaccharides in Acanthopanax sessiliflorus(Rupr.et Maxim.)were determined by Colorimetry.Rutin and Glucose were standard preparation,and wave length were 500nm and 620nm.The statistical analysis was performed with SPSS 16.0.The mass of Rutin(mg) and Abs were linear in the range of 0.05mg-0.5mg,R2=0.9996.The mass of glucose(mg) and Abs were linear in the range of 0.0208mg-0.104mg,R2=0.9996.The contents of total flavonoid and polysaccharides in leaves of biomimetic Acanthopanax sessiliflorus showed apparently higher than that in leaves of wild Acanthopanax sessiliflorus.The differences of the polysacharides and total flavonoid between wild and simulating samples are not obvious.The method was simple,quick,accurate,can be used to determine the content of total flavonoid and polysaccharides in the leaves of Acanthopanax sessiliflorus(Rupr.et Maxim).
