摘要
单克隆抗体技术是现代生命科学研究的重要工具,其在基因和蛋白质的结构与功能研究方面有着不可或缺的作用。近年来,随着分子生物学和细胞生物学的发展,出现了抗原特异性的B淋巴细胞杂交瘤技术、人-鼠嵌合抗体制备技术、噬菌体展示技术、核糖体展示技术和转基因小鼠技术制备单克隆抗体,这些技术有效地解决了单克隆抗体鼠源性等问题,使单克隆抗体凭借自身高度特异性和均一性的特点在治疗肿瘤、自身免疫性疾病和器官移植等方面显出了独特的优势和良好的应用前景,故单克隆抗体制备技术的发展也就显得尤为重要。
Monoclonal antibody technique,as an important tool in modern life science research,plays an indispensable role in the study of gene structure and protein function.Recently,with the development of molecular biology techniques and cell biology,Several methods to generate McAb have evolved,such as immortalization of antigen-specific B cell hybridoma technology,generation of chimeric and humanized antibody from antibody by genetic engineering,Phage display technology,Ribosome display technology and Transgenetic mouse,this methods can revolved the murine problem of McAb and so on.McAb has now gained a niche as an epochal breakthrough in the treatment of the tumor,autoimmune disease,organ transplanting and so on.So monoclonal antibody preparation technology development is very important.
